DSDAP (1,2-distearoyl-3-dimethylammonium-propane) is a cationic ionizable lipid with low cytotoxicity and high cellular uptake efficiency. DSDAP is neutral at physiological pH, but acquires a positive charge at low pH, such as the inside of endosomes. The electrostatic interactions between DSDAP and naturally occurring anionic lipids in endosomal membranes trigger the release of nucleic acid. DSDAP is often used in the formulation of nanocarriers (LNPs, lipid-polymer hybrid nanoparticles) used to encapsulate bioactive molecules, including mRNA, RNA, and plasmid DNA.
