DPDAP (1,2-Dipalmitoyl-3-dimethylammonium-propane) is an ionizable cationic lipid widely used in lipid-based drug delivery systems. DPDAP is neutral at physiological pH, which enhances the nanoparticle. In acidic environments, it becomes positively charged which facilitates endosomal escape and cargo release. DPDAP is often used in the formulation of nanocarriers (LNPs, lipid-polymer hybrid nanoparticles) used to encapsulate bioactive molecules, including mRNA, RNA, and plasmid DNA.
