DODMA ( 1,2-dioleyloxy-3-dimethylaminopropane) is an ionizable cationic lipid used to deliver nucleic acids for mRNA vaccines and gene therapy. DODMA is neutral at physiological pH, but acquires a positive charge at low pH, such as the inside of endosomes. Outside the cell, DODMA protects the nucleic acid from degradation. Once inside the cell, the positively charged DODMA disrupts the endosomal membrane, releasing the nucleic acid into the cytoplasm. DODMA is often used in the formulation of lipid nanoparticles (LNPs) and lipoplexes to deliver nucleic acids like mRNA and DNA.
