DMTAP (1,2-dimyristoyl-3-trimethylammonium-propane chloride) is a cationic lipid and an analogue of DOTAP. It is used for the formulation of lipid nanoparticles (LNPs) to deliver DNA, RNA, oligonucleotides, and other negatively charged biomolecules. DMTAP has saturated dimyristoyl tails (14:0), while DOTAP has unsaturated dioleoyl tails (18:1). Cationic lipids, like DMTAP, impart a positive charge to the surface of liposomes which promotes efficient interaction with negatively charged nucleic acids and the negatively charged cell membrane. Uptake of DMTAP-containing LNPs is influenced by buffer type, lipid composition of the LNP, and cell type.
