DMDAP (1,2-Dimyristoyl-3-dimethylammonium-propane) is an ionizable lipid used in the formulation of lipid nanoparticles (LNPs) for gene delivery. DMDAP is neutral at physiological pH, but acquires a positive charge at low pH, such as the inside of endosomes. The change in pH promotes stability of the LNP at neutral pH and helps release cargo inside the cells. DMDAP is often used in the formulation of nanocarriers (LNPs, lipid-polymer hybrid nanoparticles) used to encapsulate bioactive molecules, including mRNA, RNA, and plasmid DNA.
