DSTAP (1,2-distearoyl-3-trimethylammonium-propane chloride) is a cationic lipid and an analogue of DOTAP. It is used for the formulation of lipid nanoparticles (LNPs) to deliver DNA, RNA, oligonucleotides, and other negatively charged biomolecules. DSTAP has saturated distearoyl tails (18:0), while DOTAP has unsaturated dioleoyl tails (18:1). Cationic lipids, like DSTAP, impart a positive charge to the surface of liposomes which promotes efficient interaction with negatively charged nucleic acids and the negatively charged cell membrane. Uptake of DSTAP-containing LNPs is influenced by buffer type, lipid composition of the LNP, and cell type.
