DPTAP (1,2-dipalmitoyl-3-trimethylammonium-propane chloride) is a cationic lipid and an analogue of DOTAP. It is used for the formulation of lipid nanoparticles (LNPs) to deliver DNA, RNA, oligonucleotides, and other negatively charged biomolecules. DPTAP has saturated dipalmitoyl tails (16:0), while DOTAP has unsaturated dioleoyl tails (18:1). Cationic lipids, like DPTAP, impart a positive charge to the surface of liposomes which promotes efficient interaction with negatively charged nucleic acids and the negatively charged cell membrane. Uptake of DPTAP-containing LNPs is influenced by buffer type, lipid composition of the LNP, and cell type.
